Induced Ascending pH Gradients in Liquid Chromatography with Flowing Retentive Stationary Phase

Abstract

The possibility of induced increasing pH gradients formation in the liquid chromatography with flowing retentive stationary phase was shown. The tephlonic and metallic capillars (internal V = 1 ml) were used as open chromatographic columns with flowing retentive liquid phase; water-methanolic or water-acetonitilic borate buffer solutions were used as mobile phase. The octanoic and heptanoic acids or their mixture (1:1), inducing the pH gradient formation within open capillar, were simultaneously the flowing retentive liquid phase (stationary phase). The more smooth increasing pH gradients from 5,6-5,8 to 9,1 were induced in system: 5 mcl of octanoic acid, mobile phase - 20 mM Na2B4O7 in 30% CH3OH at flow rate 0,2 - 0,3 ml/min.
