Optimization of hybridization analysis on DNA-microarrays
with colorimetric detection on basis of horseradish peroxidase
Abstract
The method of hybridization analysis on DNA-microarrays
with colorimetric detection on basis of horseradish peroxidase was developed.
The effectiveness of biotin incorporation in nucleotide sequence during PCR and
the influence of different hybridization temperatures, molarity of
hybridization buffer and lengths of labeled DNA fragments on sensitivity and
specificity of hybridization signals were analyzed. The method developed was
applied for genotyping of CTX-M І-lactamases.
