Regulation of β-amylaseʼs
activity by alkylresorcinols
Abstract
Herein the effects of two alkylresorcinols
(methylresorcinol (MR) and hexylresorcinol (HR) - chemical analogous of natural
anabiosis factors of some bacteria) on the catalytic activity of β-amylase are shown. MR stimulated the activity of β-amylase up to 170% within the range of concentrations 0.8-12.9 mM
when the water-soluble potato starch as substratum was used. HR displayed weak
stimulated effect up to 115% within the range of concentrations 0.05–0.35 mM, and
then inhibition is observed up to 50% (0.75 mM). Interaction MR with β-amylase allows to increase general yield of potato and malt starch
hydrolysis products in 1.5-1.6 times at enhancing of hydrolysis rate in 4-5
times. The increasing of modified enzyme activity was demonstrated in extended
temperature and pH ranges. The kinetic parameters of starch hydrolysis by β-amylase modified with alkylresorcinols were determined. MR
demonstrated the non-competitive type of activation, HR - the non-competitive
type of inhibition.
Key words: alkylresorcinols, β-amylase, starch, malt, stimulation and inhibition of the catalytic
activity.
